Description

Background

ACE2 antibody detects human ACE2. Angiotensin-converting enzyme 2 (ACE2) plays a central role in vascular, renal, and myocardial physiology. In contrast to its homolog ACE, ACE2 expression is restricted to heart, kidney, and testis. Recently, ACE2 has also been shown to be a functional receptor of the SARS coronavirus. The normal function of ACE2 is to convert the inactive vasoconstrictor angiotensin I (AngI) to Ang1-9 and the active form AngII to Ang1-7, unlike ACE, which converts AngI to AngII. While the role of these vasoactive peptides is not well understood, lack of ACE2 expression in ace2-/ace2- mice leads to severely reduced cardiac contractility, indicating its importance in regulating heart function. Anit-ACE2 antibodies are ideal for investigators involved in enzyme and cardiovascular research.

Immunogen

ACE2 Antibody was produced from whole rabbit serum prepared by repeated immunizations with a synthetic peptide corresponding to amino acids near the C-terminus of human ACE2.

Application Note

Anti-ACE2 Antibody is suitable for use in E, WB, IF, and IHC. Expect a band approximately ~92.4 kDa on specific lysates. Western Blot validated in human, mouse and rat samples. Immunohistochemistry and Immunofluorescence validated in human samples. Specific conditions for reactivity should be optimized by the end user.

Purity/Specificity

Anti-ACE2 Antibody was affinity purified from monospecific antiserum by immunoaffinity purification. A BLAST analysis was used to suggest cross-reactivity with ACE2 with Human based on 100% homology with the immunizing sequence. Predicted species reactivity based on immunogen sequence: Bovine: (88%), Mouse: (78%), Rat: (78%). Cross-reactivity with ACE2 from other sources has not been determined. Anti-ACE2 has no cross response to ACE1.

Storage Condition

Store vial at -20° C prior to opening. Aliquot contents and freeze at -20° C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use.
