Description

Background

Angiotensin-converting enzyme 2 (ACE2) plays a central role in vascular, renal, and myocardial physiology. In contrast to its homolog ACE, ACE2 expression is restricted to heart, kidney, and testis. Recently. ACE2 has also been shown to be a functional receptor of the SARS coronavirus. The normal function of ACE2 is to convert the inactive vasoconstrictor angiotensin I (AngI) to Ang1-9 and the active form AngII to Ang1-7, unlike ACE, which converts AngI to AngII. While the role of these vasoactive peptides is not well understood, lack of ACE2 expression in ace2-/ace2- mice leads to severely reduced cardiac contractility, indicating its importance in regulating heart function.

Application Note

Anti-ACE2 Antibody has been tested for use in ELISA, Immunofluorescence, Immunohistochemistry and Western Blotting. IHC and WB validated in human, mouse, and rat samples, IF validated in human samples. Expect a band at ~93 kDa in size corresponding to ACE2 protein by Western Blotting in the appropriate cell lysate or extract. Specific conditions for reactivity should be optimized by the end user.

Purity/Specificity

Anti-ACE2 Antibody was affinity purified from monospecific antiserum by immunoaffinity chromatography. Predicted species reactivity based on immunogen sequence: Mouse: (93%), Rat: (93%), Bovine: (93%). Anti-ACE2 has no cross response to ACE1.

Storage Condition

Store vial at -20° C prior to opening. Aliquot contents and freeze at -20° C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use.
