Background

Protein-tyrosine Phosphatase H1 (PTPH1) has recently been identified as a specific p38γ MAPK phosphatase which is mediated through PDZ interaction. Ras has been demonstrated to increase both p38 gamma and PTPH1 protein expression, and there is a coupling of increased p38 gamma and PTPH1 protein expression in primary colon cancer tissues. Phosphorylation of PTPH1 at Ser459 leads to PTPH1 stabilization, which plays an important role in Ras oncogenesis and stress response. Additionally, phosphorylation of PTPH1 Ser459 reveals a novel mechanism by which MAPK signals through PTPH1 to regulate cellular response. PTPH1 pS459 Antibody is ideal for researchers interested in cancer research.

Anti-Phospho-Ser459 PTPH1 Antibody was produced in rabbits by repeated immunizations with a synthetic phospho-peptide corresponding to amino acid residues surrounding the phospho-Ser459 of human PTPH1 conjugated to KLH.
Anti-Phospho-Ser459 PTPH1 Antibody is directed against PTPH1 protein phosphorylated at S459. The antibody was prepared from monospecific antiserum by immunoaffinity chromatography using phospho peptide coupled to agarose beads followed by solid phase adsorption(s) against non-phospho peptide and non-specific peptide to remove any unwanted reactivities. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit Serum. This antibody is specific for phosphorylated PTPH1. Minimal reactivity occurs against non-phosphorylated PTPH1. Reactivity against PTPH1 occurs from human sources. However, reactivity is also expected against bovine, canine, non-human primate, and sheep based on 100% sequence homology.

Anti-Phospho-Ser459 PTPH1 Antibody is tested for Western Blots and is specific for the ~104 kDa PTPH1 protein phosphorylated at Ser459. Immunolabeling is blocked by preadsorption of the antibody with the phosphopeptide used as antigen but not by the corresponding dephosphopeptide. Immunolabeling is also completely eliminated by treatment with lambda-phosphatase. Specific conditions for reactivity should be optimized by the end user.
