Anti-Idiotypic Antibodies

Conduct critical bioanalytical and therapeutic antibody development assays with confidence by partnering with Rockland for custom anti-idiotypic antibody generation.

For more than 25 years, Rockland has supported biopharma companies and CROs with high-quality anti-idiotype (anti-ID) antibodies used in pharmacokinetic (PK), anti-drug antibody (ADA), and ligand-binding assays. Our experienced scientific team designs each anti-ID antibody generation project to align with your assay format, regulatory expectations, and development timeline, whether you require polyclonal reagents or hybridoma-derived monoclonal antibodies.

Anti-idiotype Antibody Types

Rockland generates Type I, Type II, and Type III anti-idiotypic antibodies to support a wide range of bioanalytical applications. The appropriate anti-ID type is selected based on the assay objective and the need to detect free drug, total drug, or functional binding.

Anti-ID antibodies can be generated in multiple species from our on-site vivarium, including rabbit, mouse, rat, goat, and chicken.

Types of Anti-ID Antibodies

Type I

Inhibitory / neutralizing
+
Detects free antibody drug

Type II

Not inhibitory
+
Detects total antibody drug (free, partially bound, fully bound)

Type III

Antibody drug-target complex specific
+
Detects bound antibody drug only

Anti-Idiotype Polyclonal Antibody Generation

Anti-idiotype polyclonal antibodies are often selected for early-stage programs or applications requiring robust signal and rapid turnaround. A typical anti-ID polyclonal project follows Rockland’s established polyclonal antibody generation workflow, beginning with antibody development if the therapeutic antibody target is provided. Rockland can also generate the target antibody as part of a broader program. Visit Custom Polyclonal Antibodies or the Custom Monoclonal Antibodies to learn more.

  • Polyclonal Antibody Development

During polyclonal antibody development, animals are immunized according to a client-defined or Rockland-recommended schedule. Adjuvant selection is optimized to promote a strong and specific anti-idiotypic immune response.

A pre-bleed serum sample (5 mL) is provided for in-house screening against the therapeutic antibody to assess idiotype specificity prior to production bleeds. Immune response monitoring enables informed recommendations regarding bleed timing and project progression. Upon completion of the immunization protocol, clients may elect a terminal bleed (rabbit only) or extend the study to collect additional production bleeds on a monthly basis.

  • Polyclonal Antibody Purification

Polyclonal antibodies can be purified using a range of methods selected to support assay sensitivity, background reduction, and downstream applications, including:

    • Protein A purification
    • Protein G purification
    • Protein A/G purification
    • Affinity purification
    • IgG purification
    • IgY purification
  • Polyclonal Antibody Conjugation

To support a variety of bioanalytical assay formats, Rockland offers customizable antibody conjugation services. Available conjugates include:

    • Biotin
    • Horseradish peroxidase (HRP)
    • Alkaline phosphatase
    • Fluorochrome/dyes

Anti-Idiotype Monoclonal Antibody Generation (Hybridoma-Based)

Hybridoma-derived monoclonal anti-idiotype antibodies are often preferred for late-stage development, regulated bioanalysis, and long-term assay support due to their specificity, consistency, and renewable supply. Rockland’s hybridoma-based anti-ID monoclonal antibody services are designed to deliver highly characterized clones aligned with assay performance requirements.

In addition to custom anti-idiotype antibody generation, Rockland offers comprehensive custom polyclonal and monoclonal antibody services to support broader therapeutic development programs. For rapid access to research-grade reagents, antibodies-online.com also provides a wide selection of biosimilar monoclonal antibodies.
