Description

Background

F(ab')2 Anti-Goat IgG Biotin Antibody was generated by enzymatic cleavage and subsequent separation from the Fc fragment. Because of their smaller size, F(ab)2 fragments offer several advantages over intact antibodies for use in certain immunochemical techniques and experimental applications. F(ab)2 fragments penetrate tissue samples and show better antigen recognition and signal generation in IHC. F(ab)2 fragments lack the Fc region and therefore do not bind Fc receptors which effectively lowers background staining. F(ab')2 Antibody is ideal for investigators who routinely perform flow cytometry, immunohistochemistry or IHC and other immunoassays.

Application Note

F(ab')2 Anti-Goat IgG Biotin Antibody has been tested by ELISA and is designed for immunofluorescence microscopy, fluorescence based plate assays (FLISA) and fluorescent western blotting. This product is also suitable for multiplex analysis, including multicolor imaging, utilizing various commercial platforms. This product has been assayed against 1.0 ug of Goat IgG in a standard capture ELISA using Peroxidase Conjugated Streptavidin #S000-03 and ABTS (2,2’-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) code # ABTS-100 as a substrate for 30 minutes at room temperature.  A working dilution of 1:40,000 to 1:200,000 is suggested for this product.   The following general recommendations are suggested as starting dilutions for: immunoblotting - 1:10,000, enzyme immunohistochemistry on tissue sections - 1:500 to 1:5,000, for flow cytometry and fluorescence immunohisto/cytochemistry - 1:200 to 1:1,000.  Researchers should determine optimal titers for other applications.

Purity/Specificity

This product was prepared from monospecific antiserum by immunoaffinity chromatography using Goat IgG coupled to agarose beads followed by solid phase adsorption(s) to remove any unwanted reactivities.   Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-biotin, anti-Rabbit Serum, Goat IgG and Goat Serum.  No reaction was observed against Human Serum Proteins.

Store vial at 4° C prior to restoration.   For extended storage aliquot contents and freeze at -20° C or below.  Avoid cycles of freezing and thawing.  Centrifuge product if not completely clear after standing at room temperature.  This product is stable for several weeks at 4° C as an undiluted liquid.  Dilute only prior to immediate use.
