Background

Wild-type p53 induced phosphatase 1 (WIP1) / protein phosphatase magnesium-dependent 1 delta (PPM1D), a protein identified in the p53 DNA response pathway, is a member of the PP2C family of serine/threonine protein phosphatases which selectively inactivates p38 MAPK and dephosphorylates the ATM/ATR targets, Chk1 and p53 (1,2). Studies have shown that it is overexpressed in human cancers and is involved in the regulation of multiple DNA damage signaling pathways (2,3). PPM1D deletion results in a reduction of T and B cell function and compromised cell division, rendering cells resistant to becoming cancerous and slowing tumor development (4,5).

Anti-PPM1D antibody was prepared from whole rabbit serum produced by repeated immunizations with an 18 amino acid peptide near the C-terminus of human PPM1D.
Anti-PPM1D antibody was affinity purified from monospecific antiserum by immunoaffinity chromatography. PPM1D antibody is human, mouse and rat reactive. At least two isoforms of PPM1D are known to exist; this antibody will only detect the larger isoform.

Anti-PPM1D Antibody has been tested for use in ELISA, immunohistochemistry, immunofluorescence, and Western Blotting. Specific conditions for reactivity should be optimized by the end user. Expect a band at approximately 47 kDa in Western Blots of specific cell lysates and tissues.

Store vial at -20° C prior to opening. Aliquot contents and freeze at -20° C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use.
