Background

Parkin pS101 Antibody detects Parkin pS101 protein. Parkin is an E3 ligase in the ubiquitin-proteasome system. Hereditary Parkinson’s disease is most commonly caused by mutations in the parkin gene and is characterized by the progressive loss of dopaminergic neurons and the presence of Lewy bodies in the substania nigra. Recent evidence suggests that phosphorylation of parkin at Ser101 may have an important regulatory role on its E3 ubiquitin ligase activity. Immunogen: Anti-Parkin pS101 Antibody was produced by repeated immunizations with a synthetic phospho-peptide corresponding to amino acid residues surrounding Ser101.

Purity/Specificity: Anti-Parkin pS101 antibody is directed against human Parkin protein. The antibody was prepared from monospecific antiserum by immunoaffinity chromatography using phospho peptide coupled to agarose beads followed by solid phase adsorption(s) against non-phospho peptide and non-specific peptide to remove any unwanted reactivities. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit Serum. Immunolabeling of the parkin band is absent in parkin S101 mutants. Reactivity is expected from the following species based on 100% sequence homology: bovine and non-human primates. Cross reactivity with Parkin from other species has not been determined.

Application Note: Anti-Parkin pS101 Antibody is tested for use in Western Blotting. Specific conditions for reactivity should be optimized by the end user. Expect a band of approximately 52 kDa in size corresponding to parking protein phosphorylated at Ser101 in the appropriate cell lysate or extract.
