Background

MEK2 isoform double mutant recombinant protein is modified to put alanine residues at two key activation sites, Serine 222 and Serine 226. Mitogen-activated protein kinase kinase 2, also known as MAP2K2, MEK, MEK2, MKK2, MEK-2, is an integral component of the MAP kinase cascade that regulates cell growth and differentiation. This pathway also plays a key role in synaptic plasticity in the brain. Activated MEK 1 acts as a dual specificity kinase phosphorylating both a threonine and a tyrosine residue on MAP kinase. The inactive double mutant MEK1 recombinant protein is ideal for investigators involved in Neuroscience, Cell Signaling and Cancer Research.

Purity/Specificity: MEK2 double mutant human is a recombinant protein containing a polyhistidine tag expressed in Sf9. This double mutant protein contain alanine substitutions at S222A and S226A rendering it inactive. Analysis by SDS-PAGE resulted in a pattern consistent with purified MEK2 and was estimated to be greater than 90% pure.

Application Details

Application Note: MEK2 mutant protein is suitable as a negative control protein for immunoassays using antibodies targeting the critical MEK2 phosphorylated S222 or S226 residues. For western blot use at 50 ng or less. For other assays concentration is user optimized.
