WIT-P-NC™ Culture Medium supports the expansion of human primary mammary epithelial cells for more than 20 passages without the growth arrest that is seen with other primary epithelial cell culture media. In addition, primary mammary epithelial cells grown in WIT-P-NC™ Culture Medium exhibit a more normal luminal pattern of gene expression than cells grown in other epithelial culture media. Target Details

Purity/Specificity: Found free of microbial contamination by direct culture: No growth: Pass. Mycoplasma: Negative: Pass. Growth characteristics enabled by media is in accordance with internal QC rates and values: Pass. To make WIT-P-NC Complete Medium, combine the following: 500 mL WIT Basal Medium, 15 mL WIT-P-NC Supplement, and 500 μL cholera toxin. When handling cholera toxin, or when performing cell culture using medium with cholera toxin, gloves, lab coat and eye protection should be worn. Any media spills should be cleaned immediately. Cell culture environments should be wiped down (70% ethanol and bleach) after completing work with either of these media.

Application Details

Application Note: Suitable for normal human breast epithelium, normal human prostate epithelium, transgenic mouse prostate cancer, normal mouse prostate epithelium. Useful for derivation, maintenance, and expansion of adherent and spherical cultures. This is a defined media, which is serum free, and maintains luminal cell phenotypes. Cells were grown in culture using WIT-P-NC were passaged in accordance with the Media QC SOP. Population doubling values and population doubling rates met internal QC specifications.
